2 Research Objectives, Pririties & Operating Principles We seek t imprve the lives f individuals with Fancni anemia (FA) thrugh research that fcuses n the rapid discvery and develpment f therapies r strategies that treat, cntrl, r cure FA. The Fancni Anemia Research Fund has particular interest in supprting interdisciplinary and translatinal research effrts that effectively address ne r mre f the fllwing pririties: T understand hw alteratin f the Fancni anemia genes and their prducts lead t the clinical manifestatins f Fancni anemia. T determine the causes f bne marrw failure, myeldysplasia and leukemia in individuals with Fancni anemia, and t develp strategies t prevent, treat, and cure these disrders. T define the pathgenesis f Fancni anemia-assciated cancers, and t develp strategies fr preventin, early detectin, treatment and a cure. T identify practical and practive management strategies that families and persns with Fancni anemia can use t maintain a high quality f life. T supprt the creatin f shared resurces, databases, and technlgies fr the internatinal Fancni anemia research cmmunity. Operating Principles: We fster active engagement and partnership f Fancni anemia families with clinicians and researchers in designing and pursuing ur bjectives. We invite prpsals frm cllabrative research grups and individual investigatrs that are respnsive t the Fund s research bjectives, and advance ne r mre f the abve pririties. We spnsr cnferences and sympsia with the specific gals f advancing knwledge, publicizing discveries and fstering the bradest pssible cllabratins t advance the abve bjectives. We require all grant recipients as well as cnference and sympsium attendees t make data, mdels and tls rapidly available in the public dmain. Page 2 f 7

3 Guidelines fr Submissin f Research Grant Prpsals Nte: Prpsals must be submitted as a single PDF attachment with page numbers included. Letter f Intent Submissin: A Letter f Intent (LOI) is required fr research funding cnsideratin. The LOI is limited t ne page and must include (1) Name(s) & Institutin, (2) Specific Aims, (3) Lay Abstract, (4) Ttal Funds Requested and (5) Translatable Applicability. A submitted LOI lnger than ne page will be returned. Investigatrs will be infrmed in a timely manner whether r nt they have been invited t submit a full prpsal. Only invited prpsals will be accepted fr review. See Full Grant Applicatin Submissin requirements belw. Mst grants are funded fr a ne- r tw-year perid, but ccasinally, grants beynd tw years will receive funding. In this case, the applicant must explain why the prject requires the lnger perid by separating the tasks int separate prjects fr shrter time perids, sequentially. Full Grant Applicatin Submissin: (Pt. 1) The scientific bdy f the applicatin must be n mre than eight pages and include the fllwing (a-h): a. Prpsal Cver Sheet (see template n page 6): List specific aims and use lay language in prject summary. Use space prvided. Indicate if yu prefer t exclude certain researchers frm reviewing the applicatin. We will attempt t hnr the preference but reserves the right t select reviewers that best meet the Fund s need fr a rigrus review f the applicatin. b. Research Strategy: Include scientific backgrund, preliminary studies, hyptheses, experimental apprach (verall strategy, methdlgy, and analyses t be used), expected results and benchmarks f success, strengths and weaknesses f the prject, ptential prblems and alternative strategies. c. Prvide a Statement f Relevance t the Fund s research pririties. In additin, cmment n the prject s significance and innvatin. d. If the prject invlves FA patients, indicate whether yu will cllabrate with anther center t increase the number f FA patients in the study. List the centers and crrespnding cntacts. e. Explain hw the prject culd be sustained, if apprpriate, after the requested funding perid. f. Prvide a statement detailing ther grant supprt available t yur lab, if relevant t this prject, including any pending applicatin(s). g. Prvide a statement f facilities available fr the prject at yur institutin. h. Prvide budget narrative and justificatin using the attached Prject Budget Template (see page 7). 4 Instructins t Scientific Reviewers A single scre is given t each prpsal recmmended fr future cnsideratin as an estimated numerical indicatr f its scientific merit. Reviewers use their wn standards f excellence t arrive at this estimate f scientific quality using the set f adjectival descriptrs belw. Only applicatins with significant and substantial scientific merit are scred. All ther applicatins are "Nt Recmmended fr Further Cnsideratin. Rating System: Each reviewer vtes n a scale f 1.0 t 5.0, using increments f 0.5. The best pssible scre achieved is 1.0 with 5.0 indicating the prest rating. Ratings shuld be reached and recrded independently. Applicatins nt recmmended fr further cnsideratin are nt rated. Each reviewer is expected t use his r her persnal standards f excellence in arriving at a pririty scre. Merit assessment includes such factrs as: hw the recmmended research wuld advance knwledge in the field; the technical and scientific cmpetence f the investigatrs; the degree f institutinal supprt; the capacity f facilities and resurces; and the internal rganizatin, administratin, and quality cntrl management. The fllwing questins will be asked f each grant applicatin reviewer: Apprach f Prpsed Wrk: Are the cnceptual r clinical framewrk, design, methds, and analyses adequately develped, well integrated, well-reasned, and apprpriate t the aims f the prject? Des the applicant acknwledge ptential prblem areas and cnsider alternative tactics? Innvatin: Is the prject riginal and innvative? Fr example, des the prject challenge existing paradigms r clinical practice, and address an innvative hypthesis r critical barrier t prgress in the field? Des the prject develp r emply nvel cncepts, appraches, methdlgies, tls, r technlgies fr this area? Significance: Des this study address an imprtant prblem? If the aims f the applicatin are achieved, hw will scientific knwledge r clinical practice be advanced? What will the effect f these studies be n the cncepts, methds, technlgies, treatments, services, r preventative interventins that drive this field? Qualificatins f Investigatrs: Are the investigatrs apprpriately trained and well-suited t carry ut this wrk? Is the prpsed wrk apprpriate t the experience level f the principal investigatr and ther researchers? Des the investigative team bring cmplementary and integrated expertise t the prject (if applicable)? Budget: Is the prpsed budget reasnable? 5 Grant Review Prcess The Fancni Anemia Research Fund uses a three-tier grant review prcess: Tier One: The scientific merit f an applicatin has t be established by a minimum f tw t three peer reviews. Tier Tw: The prpsal review cmmittee (PRC), a sub-cmmittee f the bard f directrs, reviews the applicatin in cnjunctin with the peer reviews and makes a funding recmmendatin t the bard f directrs. Tier Three: The bard f directrs will determine whether t apprve r deny funding based n (1) the scientific merit f the grant prpsal, (2) cnfrmity with the FA Research Fund s research pririties, and (3) budget cnsideratins. LOIs are then reviewed by the PRC fr ptential funding determinatin. The principal investigatr is either invited t submit a full grant applicatin r is infrmed that the LOI was nt recmmended fr further cnsideratin. Upn receipt, full applicatins are prcessed and assigned external (peer) reviewers. Once peer reviews have been received, each grant applicatin is sent t the PRC fr review. The PRC deliberates each applicatin and may request further infrmatin. The PRC prduces a reprt cmpiling their feedback and the peer reviews. The PRC makes funding recmmendatins t the bard f directrs. The bard f directrs hlds an fficial vte. The principal investigatr is infrmed f the funding decisin within ne week f the vte. Selected applicants will receive an acceptance f award letter fr review and signature. Page 5 f 7
