Presenttion Amyvid 800 MBq/ml: Ech ml of solution for injection contins 800 MBq of florbetpir ( 18 F) t the dte nd time of clibrtion (ToC) (rnges from 800 MBq to MBq). Multidose vils of 10 or ml. Amyvid 1900 MBq/ml: Ech ml of solution for injection contins 1900 MBq of florbetpir ( 18 F) t the dte nd ToC (rnges from 1900 MBq to MBq). Multidose vils of 10 or ml.also contins ethnol, nd sodium (up to 37mg). Uses Dignostic use only. Rdiophrmceuticl indicted for PET imging of β-myloid neuritic plque density in the brins of dult ptients with cognitive impirment who re being evluted for Alzheimer s disese (AD) nd other cuses of cognitive impirment. Amyvid should be used in conjunction with clinicl evlution. Dosge nd Administrtion A PET scn with florbetpir ( 18 F) should be requested by physicins skilled in the clinicl mngement of neurodegenertive disorders. Amyvid imges should only be interpreted by reders trined in the interprettion of PET imges with florbetpir ( 18 F). A recent co-registered computed tomogrphy (CT) scn or mgnetic resonnce (MR) imging of the ptient to get fused PET-CT or PET-MR imge is recommended in cses of uncertinty bout the loction of grey mtter nd of the grey/white mtter border in the PET scn. The recommended ctivity for n dult weighing 70 kg is 370 MBq florbetpir ( 18 F). The volume of the injection should not be less thn 1ml nd not exceed 10ml. Elderly: Dosge djustment not required. Renl nd heptic impirment: Creful considertion of the ctivity to be dministered is required since n incresed rdition exposure is possible in these ptients. The phrmcokinetics of florbetpir ( 18 F) in ptients with renl or heptic impirment hve not been chrcterised. Florbetpir ( 18 F) is excreted primrily through the heptobiliry system nd ptients with heptic impirment hve the potentil of incresed rdition exposure. Method of dministrtion: For intrvenous use. For multidose use. The ctivity of florbetpir ( 18 F) hs to be mesured with n ctivimeter (dose clibrtor) immeditely prior to injection. The dose is dministered by intrvenous bolus injection, followed by flush of sodium chloride 9mg/ml (0.9%) solution for injection to ensure full delivery of the dose. Injection of Amyvid through short intrvenous ctheter (pproximtely 4 cm or less) minimises the potentil for dsorption of the ctive substnce to the ctheter. The injection of florbetpir ( 18 F) must be intrvenous in order to void irrdition s result of locl extrvstion, s well s imging rtefcts. Imge cquisition: A 10 minute PET imge should be cquired strting pproximtely to 50 minutes fter intrvenous injection of Amyvid. Ptients should be supine with the hed positioned to centre the brin, including the cerebellum, in the PET scnner field of view. Reducing hed movement with tpe or other flexible hed restrints my be employed. Reconstruction should include ttenution correction with resulting trnsxil pixel sizes between 2.0 nd 3.0 mm. Contr-indictions Known hypersensitivity to ny ingredient. Wrnings nd Specil Precutions Individul benefit/risk justifiction: For ech ptient, the rdition exposure must be justifible by the likely benefit. The ctivity dministered should, in every cse, be s low s resonbly chievble to obtin the required dignostic informtion. Limittions of use: A positive scn does not independently estblish dignosis of AD or other cognitive disorder since neuritic plque deposition in grey mtter my be present in symptomtic elderly nd some neurodegenertive dementis (Alzheimer s disese, Lewy body dementi, Prkinson s disese, dementi). The efficcy of Amyvid for predicting development of AD or monitoring response to therpy hs not been estblished. After the procedure: Close contct with infnts nd pregnnt women should be restricted during the initil 24 hours following the injection. This medicinl product contins 10 vol % ethnol (lcohol), ie, up to 790mg per dose, equivlent to 20ml beer or 8ml wine per dose. Interctions In vitro binding studies hve not shown interference of florbetpir ( 18 F) binding to β-myloid plques in the presence of other common medicinl products tken by AD ptients. Fertility, Pregnncy, nd Lcttion The risk benefit rtio needs to be ssessed on cse by cse bsis. Driving, etc Not relevnt. Undesirble Effects Adverse rections hve been collected in clinicl studies involving 555 subjects nd 665 dministrtions of Amyvid solution for injection. No serious dverse rections relted to Amyvid dministrtion hve been reported. While they my in relity occur t lower frequencies thn indicted below, the size of the source dtbse did not llow for the ssignment of frequency ctegories lower thn the ctegory uncommon ( 1/1,000 to <1/100). Very common ( 1/10), common ( 1/100 to <1/10), uncommon ( 1/1,000 to <1/100). Common: Hedche. Uncommon: Dysgeusi, flushing, nuse, pruritus, urticri, infusion site rections including rsh, hemorrhge, irrittion, nd pin. Other Informtion Shelf life: 800 MBq/ml is 7.5 hours, 1900 MBq/ml is 10 hours, from the ToC. Ech vil is enclosed in shielded continer of pproprite thickness to minimise externl rdition exposure.
